use | 4-bromo-2, 6-difluorobenzene methanol is an organic intermediate. It has been reported that 4-bromo-2, 6-difluorobenzene methanol can be used to prepare RET inhibitors. |
Preparation | 4-bromo -2, 6-difluorobenzyl methanol can be obtained by reducing carboxyl groups from 4-bromo -3, 5-difluorobenzoic acid. Under the protection of 0 ℃ and N2, 4-bromo -3,5-difluorobenzoic acid (3g,12.658mmol,) is dissolved in THF(60mL), BH3.Me2S(25mL,250mmol,10mol/L) is slowly added, and the reaction is naturally raised to room temperature in ice bath for overnight reaction. Slowly add methanol to quench the reaction until there is no bubble generation, concentrate, and residue silica gel column chromatography (eluent PE/EA = 15/1-10/1) to obtain 2.672g of white solid, which is the target product (yield 94.65%). |
Application example | 4-bromo-2, 6-difluorobenzene methanol can be used to prepare RET inhibitors with the following structure. RET is a proto-oncogene (transfected rearrangement gene, The REarranged duration Transfection). RET receptor tyrosine kinase is an oncogene that appears in various cancers including non-small cell lung cancer (NSCLC) and medullary thyroid carcinoma (MTC). Its abnormal activation is the key driver that leads to the growth and proliferation of various solid tumors. It has been found that the occurrence of various diseases is closely related to RET gene mutations, such as NSCLC, MTC, multiple endocrine adenomatosis type 2 (multiple endocrine neoplasiatype II, MEN2), papillary thyroid carcinoma (papillary thyroidcarcinomas,PTC), Hirschsprung's disease, lung adenocarcinoma, etc. |
